Pig farming is recognized as an activity with great polluting potential. The aim was to investigate possible environmental risks of effluents from the stabilization pond (SP) and the raw effluent (RE) from the biodigestion process of swine residues, in different concentrations in the models Lactuca sativa and Allium cepa. Seeds were germinated in different dilutions, 100% (C1), 50% (C2), 25% (C3), 12.5% (C4), 6.25% (C5), 3.12% (C6), 0.78% (C7), and 0.39% (C8). Distilled water was used as the negative control (CN) and trifluralin (0.84 g/L-1) as the positive control. Germination (GR), root growth (RG), cell cycle, and oxidative stress (OS) were analyzed. To assess OS, the activity of the enzymes superoxide dismutase (SOD), catalase (CAT), and glutathione S-transferase (GST) and the quantification of glutathione (GSH) and lipid peroxidation (LPO) were analyzed. Data were submitted to ANOVA (one way), followed by the Kruskal-Wallis mean test (P ≤ 0.05). Chemical analysis showed high values of Cu, Fe, Mn, and Zn. Dilutions (C1, C2, C3 RE) and (C1 and C2 SP) inhibited GR and RG of L. sativa and A. cepa than other concentrations. The mitotic index showed a reduction in C5 (RE), C6, and C7 (SP) of L. sativa and C3 and C4 (SP) of A. cepa in relation to CN and higher frequencies of chromosomal alterations. Regarding the OS, only the concentrations of SP treatment showed statistical difference in relation to the NC: in L. sativa model, GSH at (C5 and C8) concentrations and LPO (C7); in A. cepa model, SOD (C3 and C4), GST (C4, C5 and C6), GSH (C5 and C8), and CAT (C3 and C7). The alterations in metabolism are possibly related to the metals, such as zinc and copper, observed in high amounts in the raw waste. The results allowed us to conclude that the raw and stabilization pond effluents offer environmental risks, requiring caution and monitoring in the use of these effluents.

Pesticides are compounds with several chemical or biological agents developed to potentiate the biocide action. Their use is associated with increased economic and agricultural productivity worldwide but can harm health and the environment, damaging existing biota. Clethodim is a systemic post-emergent herbicide for grasses, highly selective for cotton, coffee, onions, carrots, soybeans, etc. Therefore, this work aimed to evaluate the harmful effect of the herbicide Clethodim with the model plant Allium cepa. A series of tests were conducted to evaluate the effects of the herbicide under study. Germination tests, root growth, cell, and nucleolar cycle analysis, as well as oxidative stress assessment and histological analysis of the roots, were performed. The results indicated that the herbicide demonstrated phytotoxicity, inhibiting germination at C1 (1.92 g/L) and C3 (0.84 g/L), and root growth at all concentrations, presenting mutagenicity at C1 (1.92 g/L) and C4 (0.24 g/L), evidenced by the increased frequency of micronuclei. In addition, changes were observed in the enzymatic activity of the enzymes catalase at concentrations C1 (1.92 g/L) and C2 (0.96 g/L) and ascorbate peroxidase at concentrations C1 (1.92 g/L), C2 (0. 96 g/L), and C3 (0.48 g/L) and in cell elongation at concentrations C1 (1.92 g/L) and C3 (0.48 g/L), demonstrated in histological analyses of the root apex.

Settleable atmospheric particulate matter (SeAPM) containing a mixture of metals, including metallic nanoparticles, has increased throughout the world, and caused environmental and biota contamination. The metal bioconcentration pattern in Nile tilapia (Oreochromis niloticus) was evaluated during a 30-day exposure to 1 g L-1 SeAPM and assessed the human health risk from consuming fish fillets (muscle) based on the estimated daily intake (EDI). SeAPM was collected surrounding an iron ore processing and steel industrial complex in Vitória city (Espírito Santo, Brazil) area. Water samples were collected daily for physicochemical analyses, and every 3 days for multi-elemental analyses. Metal bioconcentrations were determined in the viscera and fillet of fish every 3 days. The elements B, Al, V, Cr, Mn, Fe, Ni, Cu, Zn, As, Se, Rb, Sr, Ag, Cd, Pb, Hg, Ba, Bi, W, Ti, Zr, Y, La, Nb, and Ce were analyzed in SeAPM, water, and fish using inductively coupled plasma mass spectrometry. The metal concentration in SeAPM-contaminated water was higher than in control water. Most metals bioconcentrated preferentially in the fish viscera, except for the Hg and Rb, which bioconcentrated mostly in the fillet. The bioconcentration pattern was Fe > Al > Mn > Pb > V > La > Ce > Y > Ni > Se > As > W > Bi in the viscera; it was higher than the controls throughout the 30-day exposure. Ti, Zr, Nb, Rb, Cd, Hg, B, and Cr showed different bioconcentration patterns. The Zn, Cu, Sr, Sn, Ag, and Ta did not differ from controls. The differences in metal bioconcentration were attributed to diverse metal bioavailability in water and the dissimilar ways fish can cope with each metal, including inefficient excretion mechanisms. The EDI calculation indicated that the consumption of the studied fish is not safe for children, because the concentrations of As, La, Zr, and Hg exceed the World Health Organization's acceptable daily intake for these elements.

The rampant use of pesticides can cause serious environmental problems. They can be contaminating surface water and groundwater, affecting the surrounding micro and macro biota. In this sense, this work aimed to evaluate the effects of a tebuconazole-based fungicide through endpoints accessed in Lactuca sativa bioassays. Germinated-seeds with roots upon 2 mm were treated with a fungicide containing Tebuconazole (TBZ) as active compound. The final concentration of TBZ in the tested solutions were 0.025 (C1); 0.05 (C2); 0.1 (C3); 0.2 (C4) and 0.4 g/L (C5). L. sativa roots were exposed for 24 h to these solutions and Petri dishes containing the treated seeds were kept in incubation chamber at 24 °C. Two positive controls (PC,) the herbicide trifluralin (0.84 mg/L) and Methanesulfonate (4 ×10-4 mol/L), were applied. Distilled water was negative control (NC). The following endpoints were analyzed: root growth (RG), cytogenotoxic potential by cell cycle analysis, induction of DNA damage through TUNEL and comet assays. The obtained data were submitted to one-way variance analysis (ANOVA) and then to Tukey or Kruskal Wallis (P < 0.05) tests. The concentrations (C1, C2, C4 and C5) affected negatively the RG of L. sativa, in comparison with the NC. The mitotic index was reduced by 25% from NC to C1 and in the rest of treatments it did not present significant modifications. However, from C3 to C5 great amount of chromosome alterations were observed, in comparison with the NC. TBZ-based fungicide also induced DNA fragmentation as measured by TUNEL and comet assays. Thus, TBZ-based fungicide in some concentrations can have phytotoxic, cytotoxic and genotoxic effects in roots and meristematic cells of L. sativa.

Biological process treatment of landfill leachate produces a significant amount of sludge, characterized by high levels of organic matter from which humic acids are known to activate several enzymes of energy metabolism, stimulating plant growth. This study aimed to characterize humic acids extracted from landfill sludge and assess the effects on plants exposed to different concentrations (0.5, 1, 2 and 4 mM C L-1) by chemical and biological analysis, to elucidate the influence of such organic material and minimize potential risks of using sludge in natura. Landfill humic acids showed high carbon and nitrogen levels, which may represent an important source of nutrients for plants. Biochemical analysis demonstrated an increase of enzyme activity, especially H+-ATPase in 2 mM C L-1 landfill humic acid. Additionally, cytogenetic alterations were observed in meristematic and F1 cells, through nuclear abnormalities and micronuclei. Multivariate statistical analysis provided integration of physical, chemical and biological data. Despite all the nutritional benefits of humic acids and their activation of plant antioxidant systems, the observed biological effects showed concerning levels of mutagenicity.

Assessment of water resources requires interdisciplinary studies that include multiple ecosystem aspects. This study evaluated the water quality of Juara Lagoon (ES, Brazil) based on physical and chemical variables, cytogenetic responses in Allium cepa and phytoplankton dynamics. Three sampling sites were defined and water samples were collected during two sampling periods. Analyses such as determination of photic zone, conductivity, and concentrations of nutrients and metals were conducted as well as cytotoxic, mutagenic, and genotoxic potentials using A. cepa test. The main attributes of phytoplankton community, such as total richness, total density, density by class, dominance, and diversity, were also evaluated. Results have revealed that Juara Lagoon has signs of artificial eutrophication at two sampling sites due to high levels of total phosphorus and ammonia nitrogen. Cytotoxic, genotoxic, and mutagenic potentials were detected as well as high concentrations of Fe and Mn. Furthermore, 165 phytoplankton taxa were recorded, with highest richness in Chlorophyceae and Cyanophyceae classes. In addition, Cyanophyceae presented as the highest density class. A. cepa test and phytoplankton community evaluation indicated that the ecological quality of Juara Lagoon is compromised.

The ethanolic extract of propolis, especially the Brazilian green type, is widely and mainly used for therapeutic purposes despite the lack of knowledge about its effects and its cellular mode of action. This type of propolis, derived from Baccharis dracunculifolia (alecrim-do-campo), has been extensively commercialized and the consumers use it to enhance health. This work aimed to assess the genotoxic/mutagenic and antigenotoxic/antimutagenic potentials of the ethanolic extracts of Brazilian green propolis and of B. dracunculifolia, on mammalian cells. It was not observed genotoxic and mutagenic effects by both extracts. After evaluate the exposure of the cells to each extract with a recognized mutagen, simultaneously, the results showed a significant reduction on DNA damage. The experiment carried out with a pre-incubation period was more effective than without incubation test, showing that the tested extracts were able to inactivate the mutagen before it could react with the DNA.

Roots of mangrove trees have an important role in depurating water and sediments by retaining metals that may accumulate in different plant tissues, affecting physiological processes and anatomy. The present study aimed to evaluate adaptive changes in root of Rhizophora mangle in response to different levels of chemical elements (metals/metalloids) in interstitial water and sediments from four neotropical mangroves in Brazil. What sets this study apart from other studies is that we not only investigate adaptive modifications in R. mangle but also changes in environments where this plant grows, evaluating correspondence between physical, chemical and biological issues by a combined set of multivariate statistical methods (pattern recognition). Thus, we looked to match changes in the environment with adaptations in plants. Multivariate statistics highlighted that the lignified periderm and the air gaps are directly related to the environmental contamination. Current results provide new evidences of root anatomical strategies to deal with contaminated environments. Multivariate statistics greatly contributes to extrapolate results from complex data matrixes obtained when analyzing environmental issues, pointing out parameters involved in environmental changes and also evidencing the adaptive response of the exposed biota.

Mangroves are dynamic environments under constant influence of anthropic contaminants. The correlation between environmental contamination levels and possible changes in the morphology of plants, evaluated by multivariate statistics helps to highlight matching between these variables. This study aimed to evaluate the uptake and translocation of metals and metalloids in roots and leaves as well as the changes induced in both anatomy and histochemistry of roots of Laguncularia racemosa inhabiting two estuaries of Espírito Santo (Brazil) with different pollution degrees. The analysis of 14 elements in interstitial water, sediments and plants followed by multivariate statistics, allowed the differentiation of studied sites, showing good match between levels of elements in the environment with the corresponding in plants. L. racemosa showed variations in their root anatomy in different collection areas, with highest values of cortex/vascular cylinder ratio, periderm thickness and air gap area in Vitória Bay, the most polluted sampling area. These three parameters were also important to differentiate the mangrove areas by linear discriminant analysis. The development stage of aerenchyma in roots reflected the oxygen availability in the water, being found a negative correlation between these variables. The combined use of chemical and biological analyses responded quite well to different pollution scenarios, matching morphological responses to physical and chemical parameters, measured at different partitions within the estuary. Thus, L. racemosa can be confirmed as a reliable sentinel plant for biomonitoring of estuaries impacted by anthropic pollution.

BACKGROUND: Banana and papaya are among the most important crops in the tropics, with a value amounting to millions of dollars per year. However, these fruits suffer significant losses due to anthracnose, a fungal disease. It is well known that certain seaweed extracts possess antifungal activity, but no published data appear to exist on the practical application of this property. In the present study, five organic Brazilian seaweed extracts were screened for their activity against banana and papaya anthracnose fungi. Furthermore, cytotoxic and mutagenic effects of the extracts were evaluated by the brine shrimp lethality assay and the Allium cepa root-tip mutagenicity test respectively, while their major components were identified by gas chromatography/mass spectrometry. RESULTS: Strong fungus-inhibitory effects of Ochtodes secundiramea and Laurencia dendroidea extracts were observed on both papaya (100 and 98% respectively) and banana (89 and 78% respectively). This impressive activity could be associated with halogenated terpenes, the major components of both extracts. Only Hypnea musciformis extract showed cytotoxic and mutagenic effects. CONCLUSION: The results of this study suggest the potential use of seaweed extracts as a source of antifungal agents with low toxicity to control anthracnose in papaya and banana during storage.

The BTEX (benzene, toluene, ethylbenzene and xylene) mixture is an environmental pollutant that has a high potential to contaminate water resources, especially groundwater. The bioremediation process by microorganisms has often been used as a tool for removing BTEX from contaminated sites. The application of biological assays is useful in evaluating the efficiency of bioremediation processes, besides identifying the toxicity of the original contaminants. It also allows identifying the effects of possible metabolites formed during the biodegradation process on test organisms. In this study, we evaluated the genotoxic and mutagenic potential of five different BTEX concentrations in rat hepatoma tissue culture (HTC) cells, using comet and micronucleus assays, before and after biodegradation. A mutagenic effect was observed for the highest concentration tested and for its respective non-biodegraded concentration. Genotoxicity was significant for all non-biodegraded concentrations and not significant for the biodegraded ones. According to our results, we can state that BTEX is mutagenic at concentrations close to its water solubility, and genotoxic even at lower concentrations, differing from some described results reported for the mixture components, when tested individually. Our results suggest a synergistic effect for the mixture and that the biodegradation process is a safe and efficient methodology to be applied at BTEX-contaminated sites.

Triorganotins are environmental contaminants, commonly used in antifouling agents for boats, that bioaccumulate and thus are found in mammals and humans due to ingestion of contaminated seafood diets. The importance of triorganotins as environmental endocrine disruptors and consequent reproductive toxicity in different animal models is well known; however, the adverse effects on reproductive cycle are less well understood. The potential reproductive toxicity of tributyltin (TBT) on regular reproductive cycling of female rats was examined. Wistar female rats (12 wk old, weighing approximately 230 g) were divided into two groups: control (vehicle, ethanol 0.4%) and tributyltin (100 ng/kg/d, 7 d/wk, for 16 d by gavage). Tributyltin significantly decreased the cycle regularity (%), duration of the reproductive cycle, the proestrus and diestrus phases, and number of epithelial cell in proestrus phase. TBT also increased the duration of metestrus and the number of cornified cells in this phase. Ovary weight and serum 17ß-estradiol levels decreased markedly, accompanied by a significant increase in progesterone levels. Histological analysis showed apoptotic cells in corpus luteum and granulosa cells layer, with cystic follicles after TBT exposure. Tributyltin also elevated number of atretic follicles and corpoa lutea. The micronucleus (MN) test, using Chinese hamster ovary cells, demonstrated a concentration-dependent mutagenic effect of TBT, and at 2.0 × 10(-2)ng/ml most of the cells were nonviable. The toxic potential of TBT over the reproductive cycle may be attributed to changes found in the ovarian weight, unbalanced levels of sexual female hormones, and number of ovarian follicles and corpora lutea.

Contaminated sites must be analyzed as a source of hazardous compounds in the ecosystem. Contaminant mobility in the environment may affect sources of surface and groundwater, elevating potential risks. This study looked at the genotoxic potential of samples from a contaminated site on the banks of the Taquari River, RS, Brazil, where potential environmental problems had been identified (pentachlorophenol, creosote and hydrosalt CCA). Samplers were installed at the site to investigate the drainage material (water and particulate soil matter) collected after significant rainfall events. Organic extracts of this drained material, sediment river samples of the Taquari River (interstitial water and sediment organic extracts) were evaluated by the Salmonella/microsome assay to detect mutagenicity and by Allium cepa bioassays (interstitial water and whole sediment samples) to detect chromosomal alterations. Positive mutagenicity results in the Salmonella/microsome assay of the material exported from the area indicate that contaminant mixtures may have drained into the Taquari River. This was confirmed by the similarity of mutagenic responses (frameshift indirect mutagens) of organic extracts from soil and river sediment exported from the main area under the influence of the contaminated site. The Allium cepa test showed significant results of cytotoxicity, mutagenic index and chromosome aberration in the area under the same influence. However, it also showed the same similarity in positive results at an upstream site, which probably meant different contaminants. Chemical compounds such as PAHs, PCF and chromium, copper and arsenic were present in the runoff of pollutants characteristically found in the area. The strategy employed using the Salmonella/microsome assay to evaluate effects of complex contaminant mixtures, together with information about the main groups of compounds present, allowed the detection of pollutant dispersion routes from the contaminated site to the Taquari River sediment.

RESUMEN

An aqueous extract of Rhizophora mangle L. bark is used as raw material in pottery making in the State of Espirito Santo, Brazil. This extract presents large quantities of tannins, compounds possessing antioxidant properties. Tannin antioxidant activity, as a plant chemical defense mechanism in the process of stabilizing free radicals, has been an incentive to studies on anti-mutagenicity. The present work aimed to evaluate possible antimutagenic activity of a R. mangle aqueous extract, using the Allium cepa test-system and micronuclear (MN) assay with blockage of cytokinesis in Chinese hamster ovary cells (CHO-K1). The Allium cepa test-system indicated antimutagenic activity against the damage induced by the mutagenic agent methyl methanesulfonate. A reduction in both MN cell frequency and chromosome breaks occurred in both the pre and post-treatment protocols. The MN testing of CHO-K1 cells revealed anti-mutagenic activity of the R. mangle extract against methyl methanesulfonate and doxorubicin in pre, simultaneous and post-treatment protocols. These results suggest the presence of phyto-constituents in the extract presenting demutagenic and bio-antimutagenic activities. Since the chemical constitution of Rhizophora mangle species presents elevated tannin content, it is highly probable that these compounds are the antimutagenic promoters themselves.

The surfactant Tween 80 is widely used in genotoxicity tests in many test systems to dilute hydrophobic substances that are being investigated. In this work, the possible genotoxic effect of Tween 80 was evaluated by chromosomic aberration test in Allium cepa. Temporary treatments were performed, in which the seeds germinated in ultrapure water were kept for 24 and 72 h in the concentrations of 1, 2, 3, 4, 5, 6, 7 e 8 per cent of Tween 80. Methane sulfonate (4x10-4 M) was used as a positive control and ultrapure water as a negative control. The roots were fixed in Carnoy and stained using Feulgen method. The slides were prepared using the common method of gentle squashing and approximately 5000 cells were analyzed to each concentration tested and to controls. The mitotic index, chromosomic aberration index and mutagenicity index did not differ statistically from the negative controls, disclosing absence of citotoxicity, genotoxicity and mutagenicity of Tween 80. Therefore, Tween 80 can be used in genotoxic tests, in temporary treatments, using the A. cepa as test organism.


O surfactante Tween 80 é bastante utilizado em testes de genotoxidade, com diversos sistemas teste, para diluir as substâncias hidrofóbicas a serem investigadas. Neste trabalho, o possível efeito genotóxico desta substância foi avaliado utilizando o teste de aberrações cromossômicas em Allium cepa. Foram realizados os tratamentos temporários, no qual as sementes, germinadas em água ultra-pura, permaneceram por 24 e 72 h, nas concentrações de 1, 2, 3, 4, 5, 6, 7 e 8 por cento de Tween 80. Metil metano sulfanato (4x10-4 M) foi utilizado como controle positivo e água ultra-pura como controle negativo. As raízes foram fixadas em Carnoy e coradas pelo método de Feulgen. As lâminas foram preparadas pelo método de esmagamento suave e aproximadamente 5000 células foram analisadas para cada concentração testada e para os controles. Não foram encontradas diferenças estatisticamente significativas entre as diferentes concentrações e o controle negativo, em relação ao índice mitótico, índice de aberrações e índice de mutagenicidade, o que revela ausência de efeito citotóxico, genotóxico e mutagênico do Tween 80.Desta forma, o Tween 80 pode ser utilizado em testes de genotoxidade, em tratamentos temporários, utilizando oorganismo teste A. cepa.

Cytotoxicity of metals is important because some metals are potential mutagens able to induce tumors in humans and experimental animals. Chromium can damage DNA in several ways, including DNA double strand breaks (DSBs) which generate chromosomal aberrations, micronucleus formation, sister chromatid exchange, formation of DNA adducts and alterations in DNA replication and transcription. In our study, water samples from three sites in the Córrego dos Bagres stream in the Franca municipality of the Brazilian state of São Paulo were subjected to the comet assay and micronucleus test using erythrocytes from the fish Oreochromis niloticus. Nuclear abnormalities of the erythrocytes included blebbed, notched and lobed nuclei, probably due to genotoxic chromium compounds. The greatest comet assay damage occurred with water from a chromium-containing tannery effluent discharge site, supporting the hypothesis that chromium residues can be genotoxic. The mutagenicity of the water samples was assessed using the onion root-tip cell assay, the most frequent chromosomal abnormalities observed being: c-metaphases, stick chromosome, chromosome breaks and losses, bridged anaphases, multipolar anaphases, and micronucleated and binucleated cells. Onion root-tip cell mutagenicity was highest for water samples containing the highest levels of chromium.
